چکیده

Introduction For Gram-negative bacteria, the region of the cell interfacing directly with the host/environment is the outer membrane. This membrane system is about 10 nm in diameter and is structurally, compositionally and functionally quite distinct from the cell’s cytoplasmic or inner membrane. Its main role is a protective one. It is composed of outer membrane proteins (OMPs; % 44%), some phospholipids ( z 13%) and carbohydrate polymers ( % 43%), and is unusual in as much as it is a fully asymmetric bilayer. The inner monolayer is composed largely of phospholipid and the acyl chains of lipoproteins, whereas the outer monolayer contains lipopolysaccharide (LPS) [ 1,21. Outer membranes generally contain a restricted number (three to eight) of major OMPs present in high copy number (50 000-750 000 copiedcell). In the model Gram-negative bacterium Escherichia coli, several of these (e.g. Braun lipoprotein, peptidoglycan-associated lipoprotein, and the ompA gene product) appear to play a structural role in anchoring the outer membrane to the underlying peptidoglycan layer [3, 41. Another major and important class of proteins is characterized by the OmpF, OmpC, PhoE and LamB porins which facilitate the passive diffusion of small hydrophilic solutes through the outer membrane [ S , 61. Other proteins, such as the iron-regulated OMPs, are involved in TonB-dependent uptake of more specific solutes [7, 81. Royal Irish Academy Medal Lectuit-e

OprF and OprL Conjugate as Vaccine Candidates against Pseudomonas aeruginosa; an in Silico Study

Introduction: Vaccine studies against Pseudomonas aeruginosa have often focused on outer membrane proteins (OPRs) due to their potent stimulation of the immune response. Using major outer membrane proteins of cell walls (mOMPs) of P. aeruginosa and other Gram-negative bacteria actively stimulate the immune system without any toxic side effects. Moreover, these antigens show immunological cross-...

New insight into the application of outer membrane vesicles of Gram-negative bacteria

This review presents a brief outline of our current knowledge of the structure and chemical composition of the outer membrane vesicles (OMVs), originating from the surface of Gram negative bacteria including their outer membrane proteins and lipopolysaccharides. Moreover, the functional roles and applications of OMVs in medical research such as OMV-based vaccines, OMV adjuvants properties, OMV ...

Construction of an Expression Plasmid (Vector) Encoding Brucella melitensis Outer Membrane Protein, a Candidate for DNA Vaccine

Background: DNA vaccination with plasmid encoding bacterial, viral, and parasitic immunogens has been shown to be an attractive method to induce efficient immune responses. Bacteria of the genus Brucella are facultative intracellular pathogens for which new and efficient vaccines are needed. Methods: To evaluate the use of a DNA immunization strategy for protection against brucellosis, a pla...

Immunogenicity of enterotoxigenic Escherichia coli outer membrane vesicles encapsulated in chitosan nanoparticles

Objective(s): Enterotoxigenic Escherichia coli (ETEC) is an important cause of diarrheal disease in humans, particularly in children under 5 years and travelers in developing countries. To our knowledge, no vaccine is licensed yet to protect against ETEC infection. Like many Gram-negative pathogens, ETEC can secrete outer membrane vesicles (OMVs). These structures contain various immunogenic vi...

Immunogenic Outer membrane Proteins (Omps) of Salmonella: Potential Candidate for sub-unit vaccine

The outer membrane is a continuous structure on the surface of Gram-negative bacteria and has particular significance as one of the potential targets for protective immunity. Recent studies have focused attention on the outer membrane proteins (OMPs) suggesting the existence of protective immunogenic components in Salmonella. Outer membrane protein (OMPs) has been reported to be immunogens for ...
